Handling procedure
Open vial.
Rehydrate the entire pellet with approximately 0.5 mL of #44 broth. Aseptically transfer the entire contents to a 5-6 mL tube of #44 broth. Additional test tubes can be inoculated by transferring 0.5 mL of the primary broth tube to these secondary broth tubes.
Use several drops of the primary broth tube to inoculate a #260 plate and/or #260 agar slant.
Incubate at 37°C for 24 hours in an atmosphere of 5% CO2.
Handling notes
Resuspending preserved culture in Trypticase Soy broth with 5% defibrinated sheep blood can be done as an alternative for growth. Additional transfers may be made into BHI broth. Shaking may improve growth. Additional transfers may be made into BHI broth. Shaking may improve growth.
