Cytogenetic Analysis:modal number = 53. This is a hypotriploid human cell line with the modal chromosome number occurring in 68% of cells counted. The polyploid cells occurred at 3.0%; eleven marker chromosomes were common to all cells, of which two were paired. The markers included: der(6)(q13), t(7p17q), del(10)(p11.23), paired der(7)t(7;?17)(p22;?q21.1) and der(16)t(16;?)(q24;?), and six others; normal Y, N13, N15, and N16 were absent; the X chromosomes were mostly paired.

Comments:This line was derived by A.F. Gazdar, H.K. Oie, J.D. Minna and associates from a lung cancer obtained from a patient prior to therapy.The patient was a smoker (60-pack-year).
The cells carry the K-ras 12 mutation, and there is a mutation in codon 191 of the p53 gene.
The cells are not able to synthesize the peptide neuromedin B (NMB) or the gastrin releasing peptide (GRP).
The cells stain positive for keratin and vimentin but are negative for neurofilament triplet protein.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing: Protocol: Remove medium, and rinse with 0.25% trypsin, 0.53 mM EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach.Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended

Preservation: Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
