Trace Metal Solution (see below)........ 10.0 ml
Distilled water to....................... 1.0 L Dissolve ingredients in the distilled water and adjust to pH 7 with 10 M NaOH. Boil, cool and dispense medium in 5 ml aliquots under O2-free nitrogen gas; stopper with butyl rubber and autoclave. Prior to inoculation, aseptically add to each tube the following anaerobically prepared sterile solutions:
10% Na2CO3 ......................... 0.15 ml
1.5% Na2S . 9H2O ................... 0.05 ml
Inoculate and incubate medium under 80% H2, 20% CO2 to provide conditions for H2-fixation. Adjust final pH, if necessary to 7.9 +/-0.1. Trace Metal.............................Solution:
